报告题目:Integrated Multifunctional Optical Nanoscopy for Single-Molecule Superresolution Imaging and Tracking and Its Application

Recently, light sheet-based super-resolution microscopy is a powerful technique for imaging biological samples due to its minimal photodamage. However, uneven light paths caused by solid-angle illumination can lead to optical distortions, limiting the accurate observation of individual molecules at the diffraction limit. In this study, we present an integrated multifunctional optical nanoscopy that combines differential interference contrast, total internal reflection, epifluorescence, a super-resolution radial fluctuation-stream module, and a wavelength-dependent light sheet. This advanced approach enables the simultaneous acquisition of multi-dimensional information, including three spatial coordinates (x, y, z), rotational dynamics (azimuth and elevation angles), and transport speed tracking of anisotropic nanoparticles. Additionally, it facilitates the super-localization of various nanoparticles within live cells.
The developed technique provides real-time monitoring of intracellular transport across different cellular regions, offering six-dimensional vector-valued non-fluorescence super-resolution imaging and tracking. Moreover, this method allows for in-depth four-dimensional sample observation at optimal angles, making it applicable to single-molecule and subcellular organelle imaging in live cells with sub-diffraction resolution, ultimately revealing intricate biological processes with unprecedented detail.
报告人简介:
Professor Kang received his B.S. and M.S. degrees in Chemistry from Kyung Hee University, Korea. He worked as a senior researcher and director of research at Hanil Pharmaceutical Co. from 1989 to 1997. He obtained his Ph.D. in Chemistry from Seoul National University in 1998. Following this, he was a postdoctoral research associate at Iowa State University and Ames Laboratory-U.S. Department of Energy under Dr. Edward S. Yeung from 1998 to 2000. He served as a senior researcher at Samsung Advanced Institute of Science and Technology (SAIT) from 2000 to 2002 and as a professor of Chemistry at Chonbuk National University from 2002 to 2010. He was a visiting professor at Iowa State University and Ames Laboratory from 2008 to 2010 and at Georgia State University from 2016 to 2017. He served as Head of the Department of Applied Chemistry at Kyung Hee University from 2012 to 2014 and as Vice President and President of the Analytical Chemistry Division of the Korean Chemical Society from 2020 to 2022 as well as a review board member of the National Research Foundation of Korea (NRF) from 2020 to 2024.
Currently, he is a full professor at the Department of Applied Chemistry, a professor at the Department of Chemistry, Graduate School, and director of the Institute of Natural Sciences at Kyung Hee University. Dr. Kang is also vice president of the Korea Biochip Society, director of the Korean Chemical Society and of the Korean Society of Analytical Sciences, and on the editorial boards of the International Journal of Advanced Chemical Technology (IJACT), Chinese Chemical Letters (CCL), Sustainability, Sensors, and Biosensors. He has published around 178 SCI papers and holds 20 patents. His research interests include developing various analytical systems and methods based on micro-/nano-separation, single-molecule detection-based super-resolution imaging, nanobio-sensors, and various nanobio-fusion technologies.
